Abstract

Aggregatibacter actinomycetemcomitans, particularly serotype b and to a lesser extent serotype c, is a pathogen associated with the etiology of aggressive periodontitis. Previous studies in this laboratory have showed that only 50% of patients with aggressive periodontitis with IgG response against LPS of A. actinomycetemcomitans have high titers of serum antibodies against omp29 surface protein. In addition, the transcription of omp29 gene was detected only in serotype b reference strains, but not the strains of serotype a of A. actinomycetemcomitans. Thus, to test the hypothesis that the exacerbated response against omp29 would have participation in the process of tissue destruction induced by A. actinomycetemcomitans, it will be determined the relative transcript levels of omp29 and omp29 expression by Western blot in clinical isolates of serotypes a, b and c of A. actinomycetemcomitans; and the effects of immune response against omp29 in experimental periodontal disease model in mice induced by A. actinomycetemcomitans, for the assessment of alveolar bone loss, levels of colonization by A. actinomycetemcomitans and the induction of cytokines / mediators of osteoclastic activity and levels of immune complexes in tissues. Moreover, it will be determined the transcriptome of JP2-like strain, by pan-genome microarray.
