Histopatologic analysis of the biocompatibility of the macrogol and propylene glycol of tri-antibiotic paste in subcutaneous tissue of rats

Abstract

A tri-antibiotic paste consisting of ciprofloxacin, minocycline, and metronidazole, with the vehicles macrogol ointment (polyethylene glycol) and propyleneglycol has been used for decontamination of young permanent teeth aiming the regeneration of dental pulp tissue. The aim of this study will be to evaluate the tissue response to implanted polyethylene tubes filled with macrogol or propylene glycol. Thirty rats will individually receive two polyethylene tubes, one filled with macrogol, and other propyleneglycol. An empty tube and one divulsion will be used as controls. The observation periods will be 7, 15, 30, 60 e 90 days. After each period of time, six animals will be killed and the tubes and surrounding tissue removed, fixed, and prepared to be analyzed in the light microscope with glycol methacrylate embedding, 3 mm serial cutting, and HE staining. Scores will be defined as follows: 0, none or few inflammatory cells, no reaction; 1, <25 cells, mild reaction; 2, 25 to 125 cells, moderate reaction; 3, >125 cells, severe reaction. Fibrous capsules will be categorized as "thin" when the thickness was < 150 ¼m and "thick" at > 150 ¼m. Necrosis and formation of calcification were recorded. The data will be statistically analyzed using Kruskal Wallis.(AU)
