Abstract

The purpose of this work is to analyze changes in protein profile in the acquired pellicle formed on enamel and dentine at different times in situ after exposure to two types of acid: lactic and citric. 216 blocks of enamels and 216 of human dentin (3X3 mm) will be used. The experiments will be conducted on four consecutive days. Each day, volunteers (n = 9) will use a mandibular device containing 6 blocks of enamel and 6 of dentin human. After the volunteers remain with the device in the oral cavity for 10 or 120 minutes in order to allow the formation of the acquired pellicle formation, the blocks will be immersed in citric acid (1%, pH 2.5) for 1 min or lactic acid (0.1 M, pH 4 , 8) or deionized water for 30 min, under stirring, at 37 ° C. Following, the pellicle will be removed with a filter paper soaked in 3% citric acid. This procedure will be repeated for two additional days and a 'pool' with the filter papers obtained from 9 volunteers for each type of substrate, sampling time and type of acid will be made. On the fourth day, the procedures will be repeated but the samples obtained from each volunteer will be kept isolated, to allow the analysis of individual variabilities among the volunteers. After extraction of proteins, they will be subjected to fractionation by ion exchange chromatography. The collected fractions will be concentrated and subjected to reverse phase liquid chromatography coupled to mass spectrometry (LC-ESI-MS/MS). The data obtained from MS / MS will be processed and submitted jointly to the program SQUEST (Bioworks Browser 3.2, Thermo-Finnigan, San Jose, CA, USA). The searches will be performed using the databases SWISS-PROT and Tremble.
