Gene expression profile in masticatory mucosa tissue of patients with history of chronic and aggressive periodontitis

Abstract

This study will evaluate the gene expression profile of tissue from masticatory mucosa, without the influence of biofilm, in patients with a history of chronic and aggressive generalized periodontitis compared to individuals without a history of periodontitis, with the aim to identify possible constitutive alterations in gene expression and their mechanisms, in order to clarify the biological events responsible for the different susceptibilities of individuals to the periodontal disease development. It will be selected patients with history of chronic and aggressive generalized periodontitis, and individuals with no history of periodontitis, with indication for dental implants placement. Implant placement surgeries will be performed at Graduate Clinic of Piracicaba Dental School- UNICAMP. The removed tissue to adapt the healing abutment will be collected and stored in RNA stabilization reagent (Invitrogen RNAlater ®). Tissue samples will be processing for RNA extraction, cDNA synthesis, for the subsequent preparation of hybridization reactions on microarrays, which will be assessed the expression of 44049 genes, covering the whole genome. Results will be analyzed to identify gene expression profiles of each group of patient, allowing the description of genes that had differential expression in each group as well as classes of genes that had differential modulation, and of possible pathways that these genes are related. The top 20 genes that had different expression between patient groups will be analyzed by real-time PCR, to confirm the results. Statistical analysis will be performed by ANOVA / Tukey to determine whether there are differences gene expression between groups.
