Abstract

Staphylococcus aureus is an important bacteria involved in both community and hospital-acquired infections. It stands out for its pathogenicity, dissemination, production of toxins and biofilm and great capacity for adaptation and resistance to antibiotics. Given the importance of S. aureus as one of the most frequently isolated microorganisms in wounds, this study aims to evaluate the genetic diversity, spread, virulence factors and antimicrobial resistance of Staphylococcus aureus isolated from samples of nasal and wounds of 17 patients treated at basic health units in the city of Botucatu, during the period of one year. Will be excluded from the study, those who submit admission or invasive procedures to less than one year. The collection of material from the wound will be held to "swab" sterile and after collecting the materials will be immediately sent to the Laboratory of Bacteriology Department of Microbiology and Immunology, Biosciences Institute, UNESP, to perform the planting and subsequent identification of microorganisms. The test of sensitivity to antimicrobial drugs will be performed by the technique of diffusion of the drug from agar discs impregnated according to the criteria recommended by the Clinical and Laboratory Standards Institute, using the following disks: oxacillin, cefoxitin, erythromycin, vancomycin, clindamycin, levofloxacin , Penicillin G, gentamicin, Sulfametazol / sulfamethoxazole, linezolid, quinupristin / dalfopristin, tigecycline and fusidic acid. It will also be carried out screening for vancomycin resistance and D test for detection of induced resistance to clindamycin. Will be held the polymerase chain reaction (PCR) for detection of genes encoding toxins and biofilm and multiplex PCR to characterize the type of staphylococcal cassette chromosome mec (SCCmec). Will be held the real-time PCR for rapid detection of mecA and Panton-Valentine Leukocidin. Typing by Gel Electrophoresis Pulsed-Field (PFGE) is performed to identify the profiles of clonal strains of S. aureus and the clusters are more frequently sequenced by Multilocus Sequence Typing (MLST) and spa typing.
