Abstract

Probiotic microorganisms are used in many foods or food supplements. Intestinal simulators have a good option for conducting studies on the intestinal microbiota. The simulator of the human microbial ecosystem (SHIME) consists of a connected series of reactor vessels that represent different parts of the human gastrointestinal tract with their respective values of pH, residence time, and volumetric capacity. The SHIME will be used to study the effect of Enterococcus faecium CRL 183 and Lactobacillus acidophilus 1014 on the native intestinal microbiota. Initially, the inoculum prepared with human feces in the three reactors is added and after two weeks stabilization is added daily for four weeks, E. faecium CRL 183 and L. acidophilus 1014 containing 108 CFU / mL in the three compartments that simulate the ascending colon, transverse and descending. After, four weeks of treatment the system will remain for two weeks without the addition of probiotics (washout period). During all experiments, weekly, the intestinal microbiota composition will be monitored on the enumeration of total aerobic and anaerobic bacteria, Enterococcus spp, Lactobacillus spp, Bifidobacterium spp, Enterobacteriaceae, and Clostridium spp. Each week will also be analyzed the short-chain fatty acids (acetate, propionate, and butyrate) and ammonia. The mean values of all parameters will be subjected to analysis of variance and Tukey test at p <0.05, using the statistical software Sigma Stat 5.0.(AU)
