Evaluation of the impact of consumption of Lactobacillus acidophilus 1014 and yacon extract of intestinal microbial diversity in wistar rats undergoing chemical carcinogenesis of colon

Abstract

The incidence and mortality due to colorectal cancer have shown a trend to increase worldwide. In Brazil, as life expectancy of the population has been increasing, neoplasia has been getting importance in morbity and mortality. Thus, it's necessary to develop new treatment strategies, such as the use of probiotics (L. acidophilus 1014) and prebiotics (yacon). The objective of this research is to evaluate the effects of consumption of probiotics and prebiotics on the gastrointestinal tract of animals suffering of colon cancer chemically caused induced. To do so, 175 male Wistar rats will be used, randomly assigned to five treatments divided into five cultures: Group I - control animals (animals who will eat only commercial feed and won't be chemically led to cancer); Group II - negative control animals (animals who will eat only commercial feed and will be led to cancer); Group III - (animals who will eat commercial feed + probiotic and will be led to cancer); Grupo IV - (animals who will eat commercial feed + prebiotic and will be led to cancer); Grupo V - (animals who will eat commercial feed + probiotic + prebiotic and will be led to cancer). The animals with chemically caused induced cancer will be inoculated with 1,2 dimetylhydrazine once a week during 8 months. Daily, an amount of 3 ml of probiotics and prebiotics will be provided to the rats for each kilo they weigh right through the same period of time. Afterwards, five of the animals belonging to each of the cultures will be monthly sacrificed. We'll analyze the intestinal microbiota of the rats suffering from colon cancer chemically induced by counting the specific microorganisms, the production of short chain fatty acids, ammonia, polyamines and polyacrylamide gel electrophoresis denaturing gradient (DGGE). The results will be submitted to variance analysis and Tukey range test, being its significance of 5%. (AU)
