Evaluation of the effects of acidulated phosphate fluoride on the effectiveness of CO2 laser in reducing demineralization of dental enamel - in situ study

Abstract

The objectives of this study will be to evaluate in situ whether the combination of topical acidulated phosphate fluoride (APF) with the irradiation of enamel with CO2 laser (›10.6 um) will enhance the of calcium fluoride (CaF2) and fluorapatite (AF) formation on the surface of previously demineralized enamel surface and evaluate whether the combination therapy will be effective in reducing the caries-like lesion progression of dental enamel of (mineral loss). Thus, a double-blind in situ study with 04 phase of 14 days each will be accomplished. In this study, 16 volunteers will use 01 intraoral palatal device containing 08 specimens each of previously demineralized smooth surface enamel (532 specimens). At each phase, each specimen will receive one of the following treatments: (1)non-fluoridated dentifrice (control), (2)non-fluoridated dentifrice + laser irradiation, (3)fluoridated dentifrice, (4)fluoridated dentifrice + laser irradiation, (5)non-fluoridated dentifrice + APF, (6)non-fluoridated dentifrice + APF + laser irradiation, (7)fluoridated dentifrice + APF, (8)fluoridated dentifrice + APF + laser irradiation To provide a high cariogenic challenge, the volunteers will be instructed to drip one drop of 20% sucrose solution onto each mesh that was above the enamel slab, 8 times per day at pre-determined times. On the 14th day of each phase, the enamel samples will be collected and sectioned longitudinally, 02 specimens of each group will be subjected to microhardness analysis and mineral loss expressed as % volume mineral by micrometer (”Z) and two specimens will be analyzed to determine the concentrations of CaF2 and FA on dental enamel. To evaluate the effects of the treatments, the dependent variables mineral loss (”Z), concentration of CaF2 and FA will be analyzed. Initially, we checked the assumptions of equal variances and normal distribution. The factors to be included in the statistical analysis will be voluntary, first, second, third and fourth phase, position of the specimen in the device and treatment. The results will be statistically analyzed in accordance with the most appropriate test for the study, with significance level of 5%. (AU)
