Abstract

As a part of the ongoing studies, this stage of the work involves carring out experimental models to the evaluation of the pharmacological activities attributed to the vouacapans derivatives obtained from Pterodon genus 6a-oxovouacapan-7²-17b-lactone e 6a-hidroxivouacapan-7²-17b-lactone, and harmicine, a ²-carboline obtained from Peganum genus, against nervous system disorders culminating with the appearance of neuropathic pain. This type of noxious stimulus is among the most difficult to diagnose and treat, because is not always possible to identify the causative agent, so it is necessary the research of new drugs capable of blocking neuropathic mediation, in order to control adequately the chronic pain arisen from this process. Aiming to learn new techniques for the study of neuropathic pain, we established collaboration with prof. Dr. Z. Jim Wang, from the College of Pharmacy at the University of Illinois at Chicago (UIC). This opportunity will work with experimental models that are closer to the clinical responses that will address the treatment of chronic neuropathic pain. It will be evaluated different neuropathic stimuli throughout rodent models: central mediation, with the excitotoxic spinal cord injury model (neuromodulators intraspinally injected); peripheral mediation with neuroma model (phantom limb pain), chronic constriction injury model (muscular compression) and the partial sciatic nerve ligation model (burning pain - causalgia). Still, the quantification of mieloperoxidase (MPO) activity enzyme related to the chronic inflammatory process. These results will permit us to demonstrate the compounds´ potential to be used on this kind of pain, which increasingly affects the worldwide population, lacking more effective and least toxic agents.
