In vitro citotoxicity evaluation of experimental zirconia against osteoblasts cells

Abstract

The aim of this in vitro study is to evaluate the behavior of osteoblasts cell against an Experimental Zirconium (EZ) compared to commercially available Zirconium (CZ) and to commercially pure Ti (TI CP). The control groups will be: culture medium (Negative Control - NC); phenol solution 1% (positive control - PC). Ten discs with 13 mm in diameter and 4 mm thick will be made for each group. Zirconia discs will be made from pressed blocks and machined in the CAD/CAM CEREC In Lab system and sintered for obtaining the final measures. Ti discs will be melting in an inert atmosphere of argon and injection vacuum pressure, and air abraded after does inclusion with aluminum oxide particles. All disks will be polished with a colloidal silica solution. Cells MC3T3E1 p5 the p15 (pre-osteoblasts) will grow on a solid surface, in the medium essential minimum, supplemented with fetal bovine serum 10 (SFB). Direct cytotoxicity tests will be carried out by placing the samples in direct contact with the cells, to observe the cell proliferation in contact with the material and evaluated by analysis of absorbance, in triplicate. Indirect cytotoxicity will be carried out with extract materials. After storage, the culture medium will be replaced by extracts of the materials, the PC will be replaced by phenol and the NC will be maintained in the medium culture. The plaques will be stored and subjected to absorbance analysis, in triplicate. The data from direct and indirect analyses will be analyzed, by ANOVA one-way, separately.(AU)
