Abstract

Studies on primary cell cultures represent important tool for the study of the toxic effects of severous dental materials in humans. The aim of this study is to isolate human cells of the follicle and the dental pulp of third molars subjected to extraction, to characterize these cells and keep them cryopreserved for testing bioactivity and cytotoxicity of new cements. For isolation of cells it will be tested two methods: explant + enzymatic treatment with trypsin and collagenase. Once the culture system of isolated cells is obtained, the same will be characterized by the use of specific markers for osteogenesis: bone morphogenic protein 7 (BMP-7) and dentin sialoprotein (DSP); and the formation of nodules of mineralization will be evaluated by the assays Alizarin Red and alkaline phosphatase. After maintaining the established cell culture, cytotoxicity assays will be performed (MTT and Trypan Blue) with the strains isolated by comparing them with two immortalized cell lines: fibroblasts (MPDL) and osteoblastic cells (SAOS-2), using the following cements experiments: 1) Portland cement (PC) 2) White MTA (Angelus), 3) + 30% CP ONB microparticulate; 4) CP + 30% nanoparticulate ONB
