Abstract

The interactions between fungi and bacteria are present in nature and has great environmental and medical importance. The proper development in vitro and in vivo models to characterize these interactions is essential for the understanding of the development of diseases and the research of new therapeutic strategies. The aim of this study is to evaluate the microbial interactions between Streptococcus mutans and Candida albicans biofilms models in vitro and in experimental models of invertebrates. At the in vitro study, will be evaluated the influence os S. mutans over the biofilm formation and filamentation capability of the C. albicans. In both tests, will be assessed the direct effects of the cells of S. mutans on C. albicans and also the indirect effects of S. mutans on C. albicans, using only the supernatant of their culture. In addition, tested the effects of S. mutans on C. albicans in different growth phases of bacterial culture (4, 6, 18 e 24h). For the study in vivo, the strains of S. mutans will be inoculated along with C. albicans in larvae of Galleria mellonella for induction of experimental infection. The effects of S. mutans on candidosis experimental analysis will be evaluated by curve by G. mellonella survival, for studies of tissue culture for UFC/mL quantification of C. albicans and histological evaluation of filamentation zone of C. albicans in the tissues of the host. The results of the tests in vitro and in UFC/mL count G. mellonella will be submitted to ANOVA and Tukey's test. The data obtained in the survival curve by G. mellonella will be analyzed by method of Log-rank. In all tests, will be considered a significance level of 5%. (AU)
