Abstract

Proteases represent one of the three largest groups of industrial enzymes and account for about 60% of the total worldwide sales of enzymes. Although proteolytic enzymes can be obtained from animals and plants, microorganisms are the preferred source for industrial applications in view of the scientific and economical advantages. Among various groups of microbes, psychrotrophs, are ideal candidates for enzymes obtention, because enzymes isolated from those organisms are active at low temperatures and stable under alkaline conditions, in presence of oxidants and detergents. Those qualities are desirable in laundry industry, and are also useful for bioremediation, food technology and molecular biology. At this point, it should be stated, that isolation of biological products in large scale needs efficient techniques which can preserve the bio-products properties at a relatively low cost. Aqueous two phase systems (ATPSs) emerged as a powerful method to solve this problem. However, some products require a high degree of purity, in those cases, the extraction by ATPS is not enough, so it is necessary to add one or more chromatographic steps. The aim of this project is to set up an efficient method of purification of the protease secreted by the psicrotroph yeast Rhodoturola mucilagionosa by aqueous two-phase systems (PEG/NaPA) and chromatographic systems, to obtain more robust proteases of industrial and biotechnological significance.
