Abstract

Sex hormones modulate various activities in cancer, since influencing the process of tumorigenesis, cell growth and metastasis. The ADAMTS's (a disintegrin and metalloproteinase with Thrombospondin Thrombospondin-adamalysin or motifs) are secreted proteases and are involved in functions such as the processing of procollagen to collagen N-proteinase cleavage of the proteoglycan matrix and angiogenesis.Our aim is to evaluate whether the expression levels of ADAMTS 1, 4 and 9 (mRNA and protein) and its subcellular localization are altered in the presence of the sex hormones estrogen, progesterone and testosterone in cell lines derived from ovarian tumors with different degrees of malignancy , and verify the presence of these hormones strains increase or decrease its invasiveness. Strains NIH-OVCAR-3 and ES-2, and proliferation steps after proper preparation, will be treated with concentrations of the hormones progesterone, estrogen or testosterone. Levels of ADAMTS (mRNA) will be analyzed by qPCR. The values derived from fluorescence obtained by qPCR are statistically analyzed with ANOVA and differences are considered statistically significant for pd 0.05. The Nives of ADAMTS (protein), obtained from the lysate total, will be analyzed by immunoblotting. The values of the area of each of the bands are subjected to the same statistical analysis described above. To identify the subcellular proteases in cell lines will be used immunofluorescence technique. To assess whether steroid hormones lead to an increase or decrease in the invasiveness of ovarian cancer lines will be a test of invasion, where cells are placed in front of each hormone individually and this will be compared to a control sample not treated with hormones.
