Molecular phylogenetics of Mesembrinellidae (Diptera: Oestroidea): inference of among-genera and among-species relationships, divergence time estimations and biogeographical aspects

Abstract

The family Calliphoridae (Diptera: Oestroidea), in its traditional definition, probably comprises a very diverse and heterogeneous paraphyletic grouping of calyptrate dipterous. One of the reasons that render Calliphoridae a non-natural group is the inclusion of Mesembrinellidae, a group with exclusively Neotropical distribution, as one of its subfamilies. Recent studies have repeatedly showed that this clade should be treated with family staus, sister to Tachinidae. The diversity, biology and phylogenetic relationships of its subordinated groups are still poorly understood, there being no molecular phylogenetic studies available so far for the clade. In this context, this project aims to contribute to the available knowledge of this Oestroidea group, with a study of (1) the diversity and biogeographical distribution of Calliphoridae in the Neotropical region; and (2) the phylogenetic relationships among the subordinated groups of Mesembrinellidae. The material for the analyses will be obtained by field collections and by request to possible collaborative groups. For the analyses, the initial repertoire of molecular markers will comprise the ITS2, COI and COII regions, with the possibility of further inclusion of additional markers. Alignments will be conducted with the MAFFT and 4SALE programs and will include consistency tests through reiteration with changes in the order of taxa in the matrix using the initial obtained topologies. Molecular phylogenetic analyses will be conducted using the maximum-parsimony, maximum-likelihood and Bayesian inference methods. Divergence time estimations will also be conducted using Bayesian inference methods. The project also includes a particular study concerning the use of secondary structure information of ribosomal RNA molecules (continuing the work developed by the proponent in his PhD) as an additional source of phylogenetic data, homology testing and alignment.
