Abstract

Since the late 1990s, an increase in sedentary behavior, in consumption of food rich in saturated, trans and omega-6 fatty acids, sugar and low in fiber have been found, as well as a decreased intake of fruits and vegetables, changes in feeding behavior and, as a consequence, increased prevalence of obesity in the population worldwide. Besides obesity, the quantity and quality of dietary fatty acids contributed to the emergence of a chronic low grade inflammation, related to the development of chronic non-communicable diseases. Studies in nutrigenetics aim to identify genetic variations that generate different metabolic responses to a certain type of dietary pattern in a population. Thus, genetic variations can interfere with the relationship between dietary fatty acids and inflammatory biomarkers. Accordingly, the present study aims to verify the association between single nucleotide polymorphisms (SNP) of adiponectin, Toll-like receptor (TLR)-4, Interleukine (IL)-1² and IL-6 genes and fatty acids intake and their effect over inflammatory biomarkers plasma concentrations in a transversal study - ISA Capital. Three hundred and three individuals will be studied, aged 20 to 59 years. The dietary data were collected by 24 hour food recall, applied in duplicate. These dietary data were transformed into energy and nutrients using the Nutrition Data System for Research (NDS-R, Minessota, USA) software. The home blood collection was performed by specialized professional, after fasting for 12 hours, with prior appointment. Out of plasma, IL-1², IL-6, IL-8, IL-10, tumoral necrosis factor (TNF)-±, IL-12p70, adiponectin, C-reactive protein, macrophage chemotactic protein (MCP)-1, soluble intracellular adhesion molecule (sICAM)-1 and soluble vascular adhesion molecule (sVCAM)-1 will be determined using the immunoassay multiplex technique. The genomic DNA will be extracted from the total blood using the salting out method and the SNP of of adiponectin (rs266729, rs17300539, rs16861209, rs1501299 e rs2241766), TLR4 (rs4986790 e rs5030728), Interleukine (IL)-1² (rs1143623, rs16944, rs1143627, rs1143634 e rs1143643) and IL-6 (rs1800795) will be genotyped by allele specific PCR.
