Abstract

The knowledge of the microbial community involved in endodontic infections through molecular techniques and its inflammatory potential pulp and periradicular tissues are important for the understanding of the pathogenesis of diseases of endodontic origin and establishment of a more effective therapy. The objectives of this study are: 1) To investigate the presence of bacterial species targeted in cases of irreversible pulpitis and pulp necrosis by Nested-PCR; 2) To quantify the infectious load present in cases of irreversible pulpitis and pulp necrosis by Checkerboard DNA-DNA Hybridization; 3) To evaluate the effectiveness of the endodontic therapy using different auxiliary chemicals in the reduction of microbial load by Checkerboard DNA-DNA Hybridization technique; 4) To quantify LPS, through the Limulus Lysate, before (C1) and after (C2) the endodontic therapy, comparing "in vivo" the effectiveness of chemo-mechanical preparation (PQM) with 2.5% NaOCl, and Chlorhexidine 2% on its reduction; 5) To evaluate the inflammatory potential of endodontic content before (C1) and after the root canal preparation with 2.5% NaOCl and 2% Chlorhexidine (C2) in macrophage cell culture regarding the production of Pro-Inflammatory Cytokines - IL-1 ², TNF-± by the enzyme-linked immunosorbent assay (ELISA). Thirty patients will be selected for endodontic treatment due to irreversible pulpitis (n = 15) or Pulpal necrosis associated with periapical lesion (n = 15). The teeth will be divided into groups according to the auxiliary chemical used in PQM. Samples for evaluation of the microbiological content, levels of endotoxin and inflammatory potential will be collected with sterile non-pyrogenic/paper points before and after Endodontic therapy. The data will be tabulated and statistically analyzed by SPSS for Windows. (AU)
