Abstract

Teak tree (Tectona grandis L. f.) is a tropical species with great economic interest currently due to its unique wood characteristics such as strength, durability and golden color. However, despite the growth in forestry fields, little is known about its biology, particularly in terms of molecular studies. Since such studies can contribute to understand wood formation, this project aims to identify and characterize genes potentially involved in the formation of lignified tissues through detailed analysis of the T. grandis transcriptome. At first, it will be analyzed the transcriptional profile of teak lignified tissues via RNAseq, subsequently it will be evaluated differential gene expression in those tissues, and finally studies will be conducted to characterize the Cinnamyl alcohol dehydrogenase (CAD) gene family, essential for catalyzing reactions in monolignols formation, final precursors of lignin. Previous results of CAD differential gene expression in teak showed an increased in transcription in branch vascular tissues, followed by trunk tissues, and inside the wood of this tree, tissue with the highest expression were heartwood, sapwood and followed by the inner core. Consequently, all the information will be analyzed with bioinformatics tools to search for differentially expressed genes important in teak wood formation with the data generated in the RNAseq, and then specific primers will be designed to assess differences in the unigenes expression by real-time PCR. The prospect of the project is extensive and aims to have the identification of important unigenes from teak transcriptome of lignified tissues, with emphasis on CAD gene family. (AU)
