Evaluation of cytotoxicity and physical properties of endodontic cements associated with amoxicillin

Abstract

A recent study showed that addition of amoxicillin to sealers inhibited the growth of E. faecalis, before and 1, 3 and 7 days after mixing. However, the addition of substances into the sealers results in alteration of the components proportion which can result in variations in the physical properties and biocompatibility. The aim of this study is to evaluate the cytotoxicity, the setting time and the flow of AH Plus and Sealapex associated with amoxicillin in the amount of 10% of the sealer`s total weight. For evaluation of cytotoxicity, L929 fibroblasts will be exposed to different concentrations of AH Plus and Sealapex, associated or not with amoxicillin in amount of 10% of the sealer`s total weight for 24 hours. After this period it will be assessed the cell viability by means of metiltetrazólio (MTT), external morphology of the cells in scanning electron microscope and cytoskeleton using fluorescent staining for actin. The setting time and flow of the sealers will be evaluated according to the standard of the International Organization for Standardization (ISO) number 6876 (2001) and the American Society of Testing Materials (ASTM) specification C266-03 (2000). The statistical analysis will be performed using a significance level of 5%. Statistical tests will be chosen according to the distribution and homoscedasticity data.
