Abstract

The epigenetic reprogramation is crucial for genome regulation during the development of germ cells, which are responsible for originating the gametogenic lineage in mammals. The demethylation in primordial germ cells is a unique key event to erase the epigenetic memory and prevent epimutations transmissions in the next generation. There is limited information on how these events are conserved in mammals, especially in dogs. The domestic dog (Canis lupus familiaris) proves to be an ideal model for the study of development in mammals despites numerous similarities with human physiology and biochemistry. The aim of this project is to study the epigenetic dinamic of embryonic germ cells since its initial development to migration and formation of genital ridge through DNA methylation and histone acetylation patterns. In a first stage, it will be held the derivation of possible canine embryonic germ cells during periods of 20, 23, 26 and 29 days of gestation. Cells will be cultured in vitro and characterized the morphology, immunophenotype, specific markers and cell viability. When evidenced the time window in which the potential germ cells will be successfully derived and grown, epigenetic analyzes of DNA methylation and histone acetylation will be performed to characterize the time-dependent epigenetic reprogramming in dogs. We expect to elucidate the mechanisms of the reprogramming of canine germ cells, leading to a greater understanding of biotechnologic possibilities of these cells. (AU)
