Germ cells are responsible for transmitting genetic material to the next generation. The defects of formation during development may cause infertility problems in both, men and women. Several studies in animals and humans have demonstrated that it is possible to culture and differentiate of germ cells in vitro. The use of primordial germ cells (PGCs) for embryonic germ cells (EGCs) derivation has been an important model to find the acquisition of pluripotency, epigenetic remodeling and differentiation into gametes. The reprogramming events that lead the human PGCs to generate hEGCs in culture are not well understood. Therefore, this proposal aims to understand the underlying mechanisms that lead to this reprogramming during the culture in vitro, as well as analyzing the germ cell parental imprinting profile. Thus, with this study we expect to elucidate the mechanisms important for hEGC reprogramming, for a greater understanding of biotechecnological potential to be used in medicine. (AU)
