Abstract

Titanium implants have been extensively used in orthopedics and dentistry, mainly as a replacement for missing teeth. Titanium is a metal implant of choice due to its high biocompatibility and corrosion resistance, as well as absence of immuneresponse. The influence of titanium superficial characteristics on cell and biomolecules behavior, as well as in its morphology has been broadly investigated, but there are still questions about the interaction between titanium and different types of cells in a microenvironment. The purpose of the present investigation is to characterize the behavior of cells from human umbilical cord in contact with a titanium nanotexturizedsurface. The cells will be obtained from umbilical cord of healthy donors and processed after the consente of the mother; cells from subintimal vein layer will be isolated encultivated em minimal essential médium. After subconfluence, these cells wil be cultred in24-well plates (n=5) on purê comercial titanium discs (cpTi) mirror polished (control group) and on nanotexturized titanium discs by means of acid etching. The parameters to be evaluated will be:The biochemical assays to be performed will be cell proliferation and viability, total protein content and alkaline phosphatase activity in addition to the detection and quantification of mineralized nodules. It will also be performed the quantitative gene expression of genes associated to adhesion and extracelular matrix by means of Real Time PCR. All the assays will be performed in triplicate the the data obtained will be analyzed with the utilization of statistical test after normality and homogeneity tests. (AU)
