Abstract

Heart failure is a major cause of mortality and morbidity in many countries. Despite the increasing evolution in drug therapy capable of modifying its natural evolution, the time course of the disease is inevitable. The definitive treatment still remains heart transplantation, which is, however, limited by the shortage of donors. Ventricular Assist Devices (VAD) can be used in this scenario as a bridge to heart transplantation, providing circulatory support while the patient waits for transplantation. Recent studies show that, besides hemodynamic stabilization, the use of VAD can result in improved cardiac contractility, which may lead to a recovery of the capacity of the heart to pump blood. This recovery depends on processes that occur at the cellular level and modify the myocyte contractility. The force and velocity of contraction of isolated cardiac myocytes before and after implantation of the VAD Incor in sheeps will be studied in this work. The methodology will also be applied in mapping the contractile function of cardiac myocytes from pediatric patients undergoing heart surgery. (AU)
