Abstract

There are evidences that sirtuins, which correspond to enzymes that remove acetyl groups and modulate several translation initiation factors, modulate the activity of translation initiation factor in eukaryotes. In our laboratory we obtained preliminary evidences that these interactions occur in Trypanosoma cruzi by co-immunoprecipitation assays. We aim to further investigate these interactions. To do this we plan:1- To demonstrate b in vitro assays the the interaction of sir2 of T. cruzi with iF2± and eIF5A and if these interactions depend on their post-translational modifications.2- To study whether the interaction of Sir2 modulates the phosphorylation of T. cruzi eiF2± in parasite extracts.3- To study if Sir2 activity and cellular distribution varies upon different stresses.4- To detect whether eiF5A and/or eiF2a are acetylated under normal or stress conditions.
