Evaluation of multi-analyte immunoassay to quantify sexual steroids in domestic animals.

Abstract

Quantifications of sexual steroids, such as estradiol and progesterone, because of their importance, have been widely used in a great number of animal´s reproduction´s researches for the study of physiology in different species. Several methods and antibodies to quantify analytes in different biological fluids have been being developed, but each analytical methodology has its advantages and limitations. Radioimmunoassay is the most widely used method because of its efficiency and applicability in several species, but it has as major disadvantage the use of radioactive material. The concerns at this area added to the requirements of national and international´s legislations about environmental issues have increased the need of development of new techniques without the use of radioactive material. This study aims to validate the quantification of serum progesterone and estradiol in cows and ewes by the xMAP-Multiplex® technology. This technology offers advantages over radioimmunoassay such as absence of radiation´s risk and simultaneous detection of hormones, which decreases the time taken to quantify them. Blood samples will be collected from the animals of each specie, on follicular and luteal stages. Serum´s hormonal concentrations will be quantified by radioimmunoassay using previously validated kits for each hormone and specie of this study and, simultaneously, the same biological samples will be quantified by xMAP-Multiplex® technology. The Milliplex kit to be used at this experiment will be evaluated by its linearity, hormonal recuperation, analytic sensibility, precision, accuracy, by the parallelism method and physiological validation. By the execution of this work, there will be the possibility to decrease environmental impact and to increase the efficiency of hormonal quantifications in animals.
