Abstract

The lignocellulosic waste has been considered as an important substrate to biological hydrogen production, since it is widely produced, but subtilized. The sugarcane bagasse (BCA) could be used as carbon source in fermentative process. However, will be necessary the BCA pretreatment to available sugar to microorganism. In this work, the BCA will be used with and without steam explosion pretreatment. The mesophilic and thermophilic cellulolytic microbial community will be from soil samples and sludge from UASB reactor. The biomass will be enriched in an anaerobic cellulolytic medium. The assays will be performed in two steps: (I) BCA in nature will be pretreated by steam explosion to verify the effect of pretreatment on hydrogen production and (II) the better condition will be used in a new step where will be evaluated the effect of temperature and yeast extract on hydrogen production. Every assay will be realized in triplicates. As the hydrogen production as the other fermentation's products will be evaluated by gas and liquid chromatography, respectively. Microbial characterization from Bacteria Domain will be performed by microscopy and tools of molecular biology, like clone and 16S rRNA sequencing. (AU)
