Abstract

Recently pharmacological agonists of Peroxisome Proliferator-Activated Receptor (PPAR-gama) and its natural agonist 15d- prostaglandin J2 (15d- PGJ2) are showing excellent results in experimental inflammatory models such as pancreatitis, sepsis and rheumatoid arthritis , and also its potent analgesic action. The 15d- PGJ2 has anti -inflammatory properties and have been reported to mediate inhibition of cell cycle progression and cytokine production in macrophages, although much of exogenously administered 15d- PGJ2 binds to serum albumin. An alternative that widely has been able to circumvent this problem is modified release through the use of drug delivery systems. The use of nanostructured drug delivery systems like liposomes, polymeric micro- and nanoparticles and solid lipid nanoparticles have been studied with the aim of improving the therapeutic properties of the drug incorporated into them , leading to numerous advantages such as increased stability of the drug, slower and prolonged release, decreased toxicity, targeting the target tissues, among others. Thus the aim of this project is to develop (to prepare and characterize) a formulation of solid lipid nanoparticles for modified release of 15d- PGJ2 as well as conduct the investigation of immunomodulatory potential of the same formulation, envisioning a new system for future use in the treatment of inflammation . Because it is a new formulation is also to evaluate them for in vitro cytotoxicity.
