Abstract

Studies have shown that most individuals trying to lose weight regains the lost weight along time. This fact impairs the treatment of obesity in the long term. Additionally, although the hormone leptin plays a key role in the control of food intake and energy balance, most obese individuals has high circulating levels of this hormone. Therefore, it is believed that obesity is characterized by a leptin resistance state. In this context, several studies have shown that a group of proteins known as suppressors of cytokine signal (SOCS) has a substantial role in the etiology of leptin resistance, because they act as signaling inhibitors of this hormone. Although it is quite clear the involvement of SOCS3 on leptin resistance caused by diet-induced obesity, no study has investigated the involvement of this protein in the hyperphagia and the body weight regain after a period of caloric restriction. Thus, the aim of this study is to investigate the role of SOCS3 as a molecular component that amplifies the hyperphagia after a period of food restriction. Therefore, mice with inactivation of Socs3 gene only in leptin receptor-expressing cells were used. These animals will be subjected to a protocol of food restriction and their food intake and weight regain will be followed during the refeeding period. Biochemical and molecular biological analyzes will be used to elucidate the role of SOCS3 in the hyperphagia observed after the period of food restriction. In addition, preliminary results indicate an important role of SOCS3 in the control of gluconeogenesis during fasting conditions which will be investigated in detail in this project. (AU)
