Biocompatibility and mineralization capacity of tricalcium silicate-based cements: in vitro and in vivo study

Abstract

The silicate-based cements have earned highlight. For this purpose, human osteoblast-like cells SAOS-2 (ATCC HTB-85) are used for analysis of cell cytotoxicity by MTT assay and bioactivity by the enzyme alkaline phosphatase activity and the presence of calcium deposits by alizarin red stain. Moreover, the tissue reaction caused by different materials in subcutaneous of sixty rat will be evaluated. Polyethylene tubes filled with materials (tricalcium silicate + 20% zirconium oxide, MTA Plus) will be implanted and empty polyethylene tubes are used as control. After periods of 7, 15, 30 and 60 days, the animals will be euthanized with anesthetic overdose and the connective tissue adjacent to implant will be removed, fixed and embedded in paraffin. Some sections of the subcutaneous tissue will be adhered to glass slides and stained with Carazzi´s hematoxylin and eosin (H & E) to perform the morphological and morphometric analysis of the number of inflammatory cells/mm2. Other sections adhered to silanized slides will be used to perform the immunohistochemical reactions for the detection of interleukin-6 and osteocalcin. The von Kossa method is performed to identify possible calcified structures in the capsule adjacent to implants. (AU)
