Abstract

The current project aims to evaluate the effect of remineralizing agents on inhibition of proteolytic activity of caries-affected dentin and hybrid layer. 79 sound human third molars will be randomized in two assays (49 teeth - assay 1; 30 teeth - assay 2). For assay one, 95 beams (0.75 x 0.75 x 5 mm) will be obtained from the mid coronal portion of 24 molars. 76 will be submitted to an in vitro demineralization process by biological method and simulating caries-affected dentin. All the beams will be distributed in 5 groups according to the surface remineralizing treatments: G1 - sound dentin; G2 - demineralized dentin (DD); G3 - DD + 0.2% NaF; G4 - DD + MI Paste containing CPP-ACP; and G5 - Curodont Repair containing the peptide P11-4. The surfaces of 15 beams (n=3) will be analyzed by SEM. The other 80 beams will be evaluated in respect to its proteolytic activity, assessing the level of C-terminal telopeptide (ICTP) released from degraded collagen at 24 h and six months (n=8). 25 longitudinal slices (200 µm) will be obtained from the mid coronal portions of 25 molars (one per tooth) according to the groups distribution. These slices will be evaluated in respect to the abundance of MMPs and CTs using Confocal Scanning Laser Immunofluorescence Microscopy. In the assay two, 30 mid coronal portions will be obtained from 30 third molars. 24 portions will be demineralized by in vitro biological method, and they will suffer remineralizing treatments (n = 3). The bond procedure will be done for all 30 mid coronal portions. A 1-mm-thick flowable composite (Filtek Z350 XT Flow, 3M ESPE) will be applied to bonded disks and light-cured for 20 sec. Bonded specimens will be cut vertically into 1-mm-thick slabs to expose the adhesive/dentin interfaces obtaining beams. They will be evaluated as the proteolytic activity of resin/dentin interface that will be assessed by in situ Zymography at 24 h and six months. Data will be analyzed qualitatively and quantitatively, and numeric data will be assessed for normality by Shapiro-Willk test to choose parametric or nonparametric tests.
