Abstract

Organic compounds present in natural resources have been used in several experimental conditions aimed to develop safe human health dyes as alternative to dyeing industries, replacing synthetic dyes, since these are precursors of mutagenic compounds. Moreover, the use of natural dyes can add value to the final product. Many phenolic compounds extracted from microorganisms are potential natural dyes and have antioxidant activity due to the number of conjugated double bonds in the molecule. Nonetheless, in addition to a safe human health dye, an eco-friendly dye is required to be used by industries, once the international scientific community is concerned about environmental health. Thus, toxicity tests to non-target organisms have been required to evaluate adverse effects to the ecosystem. Zebrafish embryos are becoming an important tool as alternative model in ecotoxicology, due to the easy maintenance and breeding. Zebrafish embryos tests allow to analyze morphological alterations, survival and gene expression in a unique assay. Thus, the present work aims to study the natural phenolic dye erythrostominone and its photodegraded extracted from microorganism, and the synthetic dye Basic Red 51, in order to investigate early compound-induced gene expression changes and biochemical alterations potentially leading to morphological effects to zebrafish embryos (D. rerio), and to compare the morphology, transcriptomics and biochemical data among natural and synthetic dyes. The toxicogenomics potential of the dyes will be performed in zebrafish embryos by microarray analysis to verify gene expression, also biochemical changes by biomarkers analysis. Finally, we will compare the morphological, transcriptomic and biochemical data of natural and synthetic dyes, and verify sub-morphological effects to improve the predictability of the test. (AU)
