Abstract

Skeletal muscle insulin resistance is a common feature in diabetic population. One target that has been given some attention is the transcriptional factor hypoxia inducible factor (HIF-1±). Our proposal was to evaluate the role of HIF-1± in insulin resistant skeletal muscle cells. The skeletal muscle cells will isolated from quadriceps muscle from Wistar rats and cultured in DMEM (Ethics protocol no. 063/2012 FMRP-USP). After differentiation, the cells will exposed to different nutrients: glucose, palmitic acid 600 ¼M, AICAR (500 ¼M) and cobalt chloride (CoCl2 - 250 ¼M). The oxygen consumption will measured using a Clark-type electrode (Hansatech Instruments). Analyses of western blotting will performed for proteins quantifying HIF-1± and and ²-actin blotting was used as an internal control. In addition, the mRNA level of HIF-1± and ²-actin will determined by qPCR-Realtime. (AU)
