Abstract

One of the critical steps in the production of cellulosic ethanol by the enzymatic route is the efficient production of glicohydrolases. In this context the objective of this project is to increase the activity of glicohydrolases produced by Trichoderma harzianum P49P11 by culture medium optimization and classic genetic improvement. The promising preliminary results obtained in the master thesis project and the short time left motivated a transfer to a phD thesis project aiming further studies and more complete results. The main methodologies are the optimization of culture media by statiscal design and mutation of the wild strain T. harzianum P49P11 with the mutagenic agents ultraviolet irradiation and Ethyl Methanesulfonate (EMS). The supernatant of the fermentations performed with the optimized media and the improved strains will be characterized by determining the proteoma using mass spectroscopy and by determining the activities of the diferente enzymes of the cellulolitic complex. Finally the conditions hydrolysis of sugarcane bagasse pretreated by steam explosion will be optimized by the best cellulolitic complex obtained. (AU)
