Importance of Shh signaling on crosstalk between fibroblast and osteoblast

Abstract

It is known that bone remodeling is a dynamic and responsible for maintaining the anatomical and functional characteristics of bone. The biomedical point of view, there is an urgent need to understand its regulation, particularly surrounding intercellular mechanisms, which can happen in a paracrine manner. Thus, the aim of this project is to investigate if pathway of Sonic Hedgehog (Shh) is involved in crosstalk between fibroblasts and osteoblasts. It is a morphogenetic pathway involved in mechanisms which ensures perfect tissue development during organogenesis and because that we believe to be involved in the regulation of osteoblast activity during tissue remodeling mechanisms. Methodologically, will be used: fibroblasts (NIH -3T3 ) and osteoblasts (MC3T3 -E1) . After storage in the incubator, cells will be plated at day 0 (D ' 0'), and after 3 days, the culture medium will be exchanged by fresh medium , but without Fetal Bovine Serum . After 24 hour incubation, the conditioned medium will be exchanged between the lines in order to check which intracellular proteins opposite behavior to external factors, sometimes produced by osteoblasts or fibroblasts. Thereafter, biological samples will be collected properly to carry out the biological tests. The Shh pathway proteins will be evaluated by western blotting. We hope that the results of this project can bring more understanding of the mechanisms that govern the differentiation of osteoblasts, serving them with new information to combat diseases that affect the tissue.
