Abstract

The aim of this study is to evaluate the tissue response and mineralization capacity of sealers SK Seal Root Canal Sealer and Sealer 26® containing calcium hydroxide compared to AH plus®. Will be used 40 Wistar rats receive subcutaneous implants were divided into groups of 10 animals for experimental periods of 7, 15, 30 and 60 days. All animals receive four implants polyethylene tubes containing the three materials to be tested and an empty tube for control. After each postoperative period, the animals will be euthanized and polyethylene tubes with the tissue that surrounds will be removed and fixed in formalin to 10%. Then the pieces shall be split in half where one of the parties will be included in glycol methacrylate for HE staining, Von Kossa staining and without sample with polarized light and the other half will be included in paraffin, which will be processed and examined by marking immunohistochemistry for tenascin and fibronectin. The histological evaluation criterion for the subcutaneous will be 0: no or few cells and no reaction; 1: less than 25 cells and mild reaction; 2: between 25 and 125 cells and moderate reaction; 3: 125 or more cells and severe reaction. Fibrous capsule will be considered fine when <150¼m thick when> 150¼m. Von Kossa staining and polarized light analysis are regarded as positive or negative. The evaluation criteria of immunostaining is: 0 = no immunostaining; 1 = low standard of immunostaining; 2 = moderate pattern of immunostaining; 3 = high standard immunostaining; 4 = extremely high standard immunostaining. The results will be analyzed by Kruskal Wallis test and Dunn (± = 0.05). (AU)
