Abstract

This study was designed to evaluate the adhesive properties in static and under flow conditions of red cells (with or without sickle hemoglobin, Hb S) to endothelial cells from patients with sickle cell disease (HbSS and HbSC). Endothelial cells will be obtained through two different techniques, a) isolation and cultivation of circulating endothelial progenitor cells (Blood outgrowth Endothelial Cells BOECs) and b) by differentiation of endothelial cells from induced pluripotent stem cells (iPS-EC). As a control, we will use BOECs of healthy individuals and patients with other hemolytic anemias (hereditary spherocytosis) and vaso-occlusive diseases (polycythemia vera). Furthermore, we will evaluate the endothelial cells expression of inflammatory and procoagulant markers. In addition, we will assess signaling pathways involved in various cellular interactions in static culture and under flow conditions. (AU)
