Abstract

Hypersensitivity to cow's milk proteins is a major cause of food allergies, which affect mainly children. Cow milk contains over 20 proteins with allergenic potential, furthermore, ±-lactoalbumin and ²-lactoglobulin are the most important, especially the former, as it is not present in human milk. Several studies report that partial hydrolysis of these proteins reduces their allergenic potential, but none hydrolyzes proteins directly in the milk. The use of enzymes covalent immobilized to insoluble supports enables reuse and easy recovery of the product, reducing process costs. The Labenz group, led by professors, Raquel L. C. Giordano and Paul W. Tardioli, has extensive experience in the milk protein hydrolysis process using immobilized enzymes. In this scenario, this study aims to investigate the technical feasibility of generating milk with reduced allergenic potential, hydrolyzing the potential allergenic proteins, preserving its physical and organoleptic characteristics and qualities suitable for consumption, using commercial endoprotease New Pro-D® immobilized on agarose. Preliminary tests with soluble enzyme showed milk production with reduction in the proteins molecular weight and maintenance of its visual characteristics, what demonstrate this project viability. This study aims to generate products with specific sizes of peptides to be submitted to tests in vivo. If the allergenicity reduction is confirmed by the protein hydrolysis, new procedures might be studied using other endoproteases and immobilization protocols.
