Evaluation of extracelular matrix proteins during the alveolar bone healing process in ovariectomized rats treated with genisteine: microtomographical and immunohistochemical analysis

Abstract

The aim of this study is to characterize the bone tissue formed during the alveolar healing process in ovariectomized rats treated with genisteine. To perform this Project, 36 female rats with 3 months age, weighing around 200g will be divided in three experimental groups (Sham/Sal; OVX/Sal; OVX/Gen). Initially, the animals will be submited to ovariectomy or sham surgery. After 60 days, the rats of OVX/Sal or OVX/Gen will receive daily saline or medical treatment with genistein (1mg/kg/day) through gavage. These treatment wil be kept until the euthanasia moment. The right upper extraction will be performed 30 days after the beggining of the treatment and the period of evaluation of the alveolar healing process will be at 42 days after the extraction. After the eutanasia of the animals, for each experimental group (n=12), the right maxila will be removed and submited to the evaluation, considering that 6 maxilla will be separated for immunohistochemical analysis and the remaining 6 maxilla will be separated for microtomographycal evaluation.. The first analysis will have the aim to characterize the bone resorption through the imunolabeling for Osteoprotegerin (OPG), RANKL and TRAP as well as the boné formation responses, through theosteocalcin (OCN) immunolabeling). The second analysis wil be performed in order to allow the data about the bone tissue formed during the alveolar boné repair. The quantitative data will be submited to normality curve in order to deterinate the parametrycal or non parametrycal test, considering the significance level of 5%.
