Abstract

The aim of the present study is to evaluate the effect of sodium trimetaphosphate (TMP), associated or not with fluoride (F), on the concentrations of F, calcium (Ca) and phosphate (Pi) in mixed biofilms of S. mutans and C. albicans (total biofilm and biofilm fluid) formed in vitro. In addition, the number of cultivable cells, total biofilm biomass, metabolic activity of the cells, and extracellular matrix composition will be determined. TMP solutions will be prepared in the concentrations of 0.25, 0.5 and 1%, with or without F (500 ppm F). Solutions containing 500 and 1100 ppm F will be used as positive controls, while artificial saliva will be considered as negative control. Then, S. mutans ATCC 25175 and C. albicans ATCC 10231 will be cultivated for obtaining the concentrations of 108 and 107 células/mL, respectively, in artificial saliva. The lowest concentrations of TMP capable of inhibiting the growth of S. mutans and C. albicans after 24 hours will also be determined. All assays will be performed in triplicate on three different occasions. The biofilms formed on the bottom of wells of microtiter plates will be treated twice daily for 1 minute and once again on the next day. After this period, the effect of phosphates will be quantified by counting the number of cultivable cells, total biofilm biomass, metabolic activity of the cells, and extracellular matrix composition in terms of proteins and carbohydrates. Moreover, after treatment period, F, Ca and Pi in whole biofilm and in biofilm fluid will be quantified. Data will be analyzed as to its distribution (normality and homogeneity) before the determination of the appropriate statistical tests. If parametric tests are allowed, the data will be analyzed by ANOVA and Student-Newman-Keuls test. Otherwise, the Kruskal-Wallis and Dunn tests will be used. Linear regression analysis will be held in order to verify the relationship between Ca, Pi and F in the analyzed samples. The significance level adopted will be 5%.
