Proteomic analysis of the jejunum and ileum in rats exposed to chronic or acute doses of fluoride.

Abstract

Because the gastrointestinal tract is regarded as the primary route of exposure to fluoride (F), which is rapidly absorbed from it, it is believed that exposure to this element can lead to significant changes in the intestinal morphology, which can affect its function. In a previous work by our research group it was observed that exposure to F significantly interferes with the expression of several proteins of the duodenum. In addition, exposure to 50 mg/L F caused a significant decrease in the density of the overall population of neurons of the jejunum and ileum and density of immunoreactive neurons nNOS in jejunum. Thus, this study aims to evaluate the effect of exposure to acute or chronic doses of F in the proteomic profile of the ileum and jejunum. Male Wistar rats (60 days old) will be treated for 30 days with chronic doses of 0 mg/L F, 10 mg/L F or 50 mg/L F, while the acute dose will be 0 or 25mgF/kg (gastric gavage). After the experimental period, the animals will be anesthetized with 10 mg/kg body weight Anasedan + 100mg/kg of ketamine hydrochloride (Vetaset) for the collection of the ileum and jejunum. Proteomic analysis of the jejunum and ileum will be conducted in order that we can better understand the mechanisms involved in acute and chronic intoxication F by, which lead to the morphological changes observed in the previous study of our research group.
