Effects of paradoxical sleep deprivation on insulin receptor signaling and GLUT-4 in gastrocnemmius of mice

Abstract

Sleep deprivation is a present in modern society phenomenon. It is suggested that the sharp decline in the number of hours of sleep per night is related to many diseases such as diabetes and obesity. In recent years, many studies have associated reduction in total sleep time and increase in cases of obesity and diabetes type 2. This study aims to quantify the immunoreactivity and the locatlization of the insulin receptor and glucose transport protein type 4 (GLUT4), insulin-receptor substrate 1 (IRS-1) and total phosphorylated and protein kinase B (AKT) total and phosphorylated in the gastrocnemius muscle of mice. The animals will be divided into two control groups (CTL) and Paradoxical sleep Deprivation for 72 h (PSD 72h). Two experiments will be conducted separately. The first 16 animals will be divided into two groups of 8 animals, and PSP CTL 72h. In the second experiment, 32 animals will be divided into 4 groups of 8 animals, which will be assessed fasting glucose and intraperitoneal glucose tolerance test. Thus, the groups will be: 8 animals CTL + saline, 8 animals CTL + Glu, 8 animals PSP sal and 72h + 72h + 8 animals PSP Glu. After euthanasia the gastrocnemius muscles will be removed from the hind legs, frozen in liquid nitrogen and cut into cryostat for immunohistochemistry assays and Western blot.
