Abstract

Sleep is an important factor to health, it acting in processes of restoration and conservation of energy, memory consolidation, functioning of the immune system and cardiovascular health. However, animal models suggest that changes in sleep caused by deprivation and sleep restriction, are associated with considerable decreases in specific hormonal axis, promote increases of corticosterone levels, reduces in testosterone, GH, and IGF-1 and increase on thyroid hormones. This catabolic scenario establishes a set of changes that can cause a number of physiological damage, in particular in the skeletal muscle bring forty muscle atrophy condition. The mechanisms involved in muscle atrophy are still not fully understood, advances have been produced demonstrating that the ubiquitin - proteasome system and autophagy pathways are envolved, however, the role of the other markers that would justify the increase in metabolism with Sleep debt are still understood . Among the candidates, the Irisin and PGC1-± protein has gained prominence, in part; both markers are involved with body's metabolism. Thus, the aim of this study is to evaluate the effects of the paradoxical sleep deprivation and restriction, on irisin levels of rats. For this, 30 male Wistar rats, 3 months old will be distributed in three groups: a) control group (CTRL); b) Paradoxical Sleep deprivation for for 96 consecutive hours (PSP96); c) Paradoxical Sleep restriction for 21 consecutive days (RSP21D). During the experiment protocol, the control group will remain in their housing boxes in the same experimental environment. After the protocol period, the groups will be euthanized by decapitation, and blood will be collected to perform Irisina, testosterone, IGF-1, glucose, insulin, corticosterone levels, and Plantaris muscle will be carefully removed histomorphometric (cross-sectional area) and molecular analyzes (Western Blotting for PGC1-± protein). The study was approved by the Ethics Committee of the Federal University of São Paulo/Hospital São Paulo, No. 0764/10 and will be held in partnership with the Department of Psychobiology of UNIFESP.
