Abstract

This project aimed to evaluate the activity of digestive enzymes of Nile tilapia (Oreochromis niloticus) fed diets containing 0.08% of commercial probiotic AQUA-PHOTO which is composed by Lactobacillus plantarum (1.51x106 UFC/g) and Bacillus subtilis (1.34x107 UFC/g). The experimental design was completely randomized with two treatments and 48 replicates (each fish will be considered as one replicate). The 96 sex reversed Nile tilapias (mean weight of 8,18 g ± 1,85g) were distributed in 12 aquaria with volume capacity of 60 L (six aquaria received control diet and other six received diet with probiotic). At the end of seven days of adaptation with experimental diets, intestine samples (anterior, medial and posterior portions) were collected at 0, 1,2,3,4,5,6, and 7 hours after feeding. The digestive enzymatic activities of amylase, lipase, alkaline protease and alkaline phosphatase will be applied on intestine samples. For these enzymes extraction, the intestine samples will be homogenized in phosphate buffer solution (PBS), centrifuged at velocity of 13.000 xg during three minutes at 4°C and supernatant will be used to determine the enzymatic activity. The amylase and alkaline protease analysis will follow the Bernfeld (1950) and Walter (1984) modified methodologies, respectively. The lipase and alkaline phosphatase activities will be determined by specific kits (BioclinTM). Additionally, analysis of intestinal microbiota will be done by DGGE (Denaturating Gradient Gel Electrophoresis) method in Microbiology Laboratory, Malaga University (UMA) -Spain. The results will be statistically evaluated by ANOVA (variance analysis) and means will be compared by Tukey Test (p<0.05). We expected beneficial effects of probiotic added on Nile tilapia feed in order to improve the digestive enzymes activities and possible alteration of intestine microbiota. (AU)
