Effect of fluoride from dentifrice on the reduction of demineralization of fluorotic enamel

Abstract

It is still discussed if fluoride incorporated in enamel during its formation (pre-eruptive) could strength fluorotic enamel against caries. On other hand, we have recently shown in vitro that dental enamel with mild/moderate fluorosis is less resistant to demineralization than sound enamel. However, in this work the recognized local effect of fluoride (post-eruptive) of daily ingestion of fluoridated water or brushing teeth with fluoridated dentifrices was not simulated. Therefore, the aim of the present study is to evaluate if the fluoride from dentifrices could compensate greater susceptibility of fluorotic enamel to demineralization. Thus, an in vitro study with factorial design will be carried out and the factors under evaluation will be: Fluorosis in five levels, (TF 0, 1, 2, 3 and 4, classified by Thyustrup and Fejerskov index) and dentifrices in two levels, non-fluoride (placebo) and fluoridated (1,100 µg F/g as NaF). Twenty human teeth from each TF will be use. From each tooth, 2 enamel slabs will be obtained (4x3x2 mm), which will be paired within the two treatments. Half of the anatomic surface of the slabs will be protected with nail varnish to determine the baseline values of hypomineralization area and fluoride concentration. The slabs will be submitted to a pH cycling model simulating a high cariogenic challenge, and twice a day they will be treated with one of the dentifrices. At the end of 10 days of pH cycling regimen, the area of caries lesion and enamel fluoride concentration will be determined. We have hypothesized that fluoride from dentifrice should cancel the higher susceptibility of fluorotic teeth to the caries process. (AU)
