Abstract

Benign Prostatic Hyperplasia (BPH) is the disease of the highest incidence in senile dogs and the conservative treatment for dogs with reproductive interest is the use of the drug Finasteride. In the senility, the use of reproduction biotechnologies is frequent, aiming at the conservation of genetic material of dogs affected by concomitant diseases. The present study aims to compare seminal cryopreservation among dogs treated with finasteride and free from HPB. To do this, dogs of different breeds and sizes, over 6 years old, will be selected, which will constitute three experimental groups: control group (dogs free from the disease), HPB group (dogs affected by the disease) and group HPB + FIN (dogs Treated with finasteride), subdivided into two subgroups, Cryopreserved and Fresh. Seminal samples will be subjected to immediate assessments, including volume, color, appearance, motility, vigor and sperm concentration. Afterwards, morph functional evaluations will be performed: sperm morphology, semen computerized analysis (SEM), analysis of sperm DNA fragmentation by stained smear, mitochondrial activity by cytochemical staining, analysis of plasma membrane integrity (vital staining) and acrossomal staining Fast Green / Rose Bengal), lipid oxidative stress, fluorescence probes analysis through flow cytometry and sperm penetration test on perivitelin membrane in chicken egg yolk. The semen will be cryopreserved in standard diluent, adopting single step protocol. After thawing, the samples will be subjected to the same analyzes of fresh semen. The data will be evaluated by ANOVA and the groups compared by means of the T-Test, adopting a level of significance of 5%. (AU)
