Abstract

Anurans are vulnerable to various pollutants. Benzo[alpha]pyrene (BaP) is a contaminant with hepatotoxic, immunotoxic and genotoxic properties. Metabolized by the enzyme P450 (CYP1A1) produces toxic subproducts that can lead to the death of organisms. However, defense cells aid in the detoxification process. Melanomacrophages (MMs) are phagocytic cells that produce melanin, and the melanin granules can aggregate / disperse through the cytoskeleton (microtubules and actin filaments). According to the hypothesis that BaP is genotoxic, affects melanin production, microtubules and phagocytosis, our objectives will be: to evaluate the dispersion of the melanin area of hepatic MMs; Quantify the melanin production of MMs; To evaluate phagocytic activity of MMs, BaP changes in the cytoskeleton; Evaluate genotoxic effects. According to the hypothesis that ±-naphthoflavone (Anf), a CYP inhibitor, reduces subproducts of BaP, we aim to analyze how Anf acts, performing the same analyzes performed with BaP. The times will be 48 hours and 7 days, the doses will be 2mg / kg BaP and 20mg / kg Anf. There will be experiments with BaP; Tripan blue + BaP; Anf + BaP; Tripan blue + Anf + BaP. For the histological analyzes the procedures for inclusion in historesin and light microscopy analysis of the area of melanin and phagocytosis (tripan blue) will be followed. The amount of melanin will be measured by ELISA. For microtubules, pellets with MMs will be analyzed under a fluorescence microscope. For nuclear abnormalities the blood will be extracted and the procedures for analysis will be followed. (AU)
