Action of Polymyxin B and calcium hydroxide on endotoxins and microbial load of the root canal: in vivo study

Abstract

Microorganisms and their products are the main causes of pulpal pathologies and apical inflammatory lesions. During endodontic treatment, the biomechanical preparation (PBM) is not able to eliminate microorganisms, requiring the use of intracanal drugs to obtain intracanal asepsis and to aid in the modulation of inflammation. The aim of this study was to evaluate and compare the efficacy of calcium hydroxide [Ca (OH) 2] with serum or associated with chlorhexidine and Polimixin B, as intracanal medications, in channel decontamination and in reducing endotoxin levels in Primary endodontic infection and the presence of apical periodontitis, thus contributing to the daily clinical practice of endodontists. For this study, 36 teeth with primary endodontic infection were randomly divided into 3 groups: Group Ca (OH) 2: Physiological Serum + Ca (OH) 2 (n = 12), PMX Group: 10.000 IU / mL polymyxin (n = 12) and CLX Group: Chlorhexidine + Ca (OH) 2 (n = 12). Microbiological and endotoxin collections of the root canals using sterile / pyrogenic paper cone at different operative moments will be performed: Initial (S1), after PBM (S2) and after 14 days of intracanal medication (S3). Microbial culture will be performed to determine the bacterial load in CFU / mL (Colonic Forming Units / mL) present in the root canals. For quantification of endotoxins will be used the KQCL test of "Limulus Amebocyte Lysate". For statistical analysis the data will be tabulated (AU)
