The effect of association of aspirin and omega 3 in a CFA-induced inflammatory process in the temporomandibular joint (TMD) of rats: a pilot study

Abstract

Temporomandibular disorders (TMD) encompass a set of craniofacial changes, which may involve TMJ, chewing muscles and associated structures. Unlike other cyclooxygenase inhibitors, acetylsalicylic acid (ASA) increases the formation of lipoxins derived from arachidonic acid, which help in the resolution of inflammation. Resolvins derived from polyunsaturated fatty acids, especially eicosapentaenoic type 3 (Omega-3 or É-3), are catalyzed by AAS and have anti-inflammatory action in cardiovascular diseases and rheumatoid arthritis. The objectives of this study are to evaluate morphological and histochemical aspects as well as the level of cytokines in TMJ tissues in animals with Freund's complete adjuvant (CFA) + bovine collagen type II (CII) -induced arthritis. 24 adult male rats distributed in the following groups will be used: G1 - 100 ¼L of CFA + CII at the base of the tail; G2 - 100 ¼l injection of CFA + CII at the base of the tail, with daily intragastric administration of 300 mg / kg of É-3, for 7 days; G3 - 100 ¼l injection of CFA + CII at the base of the tail, with daily intragastric administration of 300 mg / kg É-3 + acetylsalicylic acid 20 mg / kg / day i.p. For 7 days. Groups 2 and 3 will be treated after 15 days of arthritis induction. The animals will be killed after seven days of treatment for both groups and ATM on both sides will be removed. An ATM will be submitted, after fixation and decalcification, to histological processing (HE, toluidine blue and red Picrossirius-hematoxylin). The contralateral TMJ will be subjected to maceration and centrifugation in buffer solution (50 mM tris-HCl, pH 7.5). The supernatant will be used for the immunoassay (ELISA) to evaluate IL1-alpha, TNF-alpha and IL-10 cytokines. The reading will be made by spectrophotometry. The results will be evaluated by ANOVA or Kruskal-Wallis test, depending on the data distribution and the scapular of the variances. Key words.
