Abstract

The design of a multifunctional polymeric system based on nanocarriers carried in microparticles for the oral administration represents a rational and promising strategy for the target of antitumor drugs to the colon. The proposed system in this project was drawn from the association of polymers previously studied by our research group and by the candidate, which add favourable properties to the desired performance, such as mucoadhesiveness, pH-responsive solubility and swelling, specific enzymatic degradability and potential of for drug target to tumour cells. Gellan gum/pectin microparticles or gellan gum/starch will be obtained and the critical attributes for their potential application will be determined. Nanoparticles based on chitosan and hydroxypropylmethylcellulose phthalate with or without hyaluronic acid and the drug camptothecin will be developed, evaluated and incorporated into the microparticles, which will be characterized according to size, shape, morphology as well as their structural characteristics. The critical attributes for the performance of the multifunctional system such as mucoadhesiveness and mucopenetration, in vitro drug release profile, in vitro intestinal permeability and antiangiogenic activity, will be evaluated. The set of data obtained will not only allow to the determination of the system potential for the application in the colorectal cancer treatment, but also contribute to the understanding of the mechanisms involved in the formation and interaction of the system in the biointerface.
