Abstract

Neste projeto de pesquisa, nós propomos investigar uma nova abordagem de coterapia, usando smart drugdelivery systems na terapia do melanoma. Essa abordagem utilizará multi-drug delivery, a partir denanoestruturas do tipo vesículas poliméricas (polimerossomos - Ps), o que representa uma alternativainovadora para o tratamento do câncer. Especificamente, o vemurafenibe, um inibidor específico daproteína B-Raf mutada no sítio V600E, e doxorrubicina, um fármaco de amplo espectro, que pode atuar emproteínas B-Raf selvagem (não mutadas), correspondem a dois importantes fármacos para a terapia docâncer tipo melanoma. A coencapsulação desses fármacos em Ps pode não apenas favorecer a estabilidadedas moléculas, mas também aumentar a meia-vida dessas moléculas na corrente sanguínea, além deapresentar possível efeito sinérgico nas células tumorais. Paralelamente a essa abordagem, propomos o usode estruturas tridimensionais (3-D) de hidrogéis de quitosana como plataformas de Ps. Espera-se que essaplataforma tenha vantagens, tais como mucoadesividade, biocompatibilidade e liberação controlada esustentada, o que a torna potencialmente interessante na aplicação transdérmica para drug delivery. Alémdisso, ao decorar a superfície dessas nanoestruturas com um ligante de afinidade tal como a transferrina(Tf), poderemos aumentar a especificidade dos Ps em relação às células tumorais e conseguir maior eficáciaterapêutica e diminuição da toxicidade. Neste trabalho, serão investigados: (i) desenvolvimento de Ps emdiferentes condições; (ii) encapsulamento (individual e simultâneo) de fármacos; (iii) decoração dos Ps comTf; (iv) caracterização das formulações dos Ps; (v) estabilidade e incorporação dos Ps em hidrogéis dequitosana sob diferentes condições; (vi) estabilidade do hidrogel com e sem Ps; (vii) liberação dos Psincorporados no hidrogel em condições simulando a via transdérmica; e (viii) comparação do efeitocitotóxico em células tumorais (BRAF mutadas ou não) versus células normais.
